Fenspirid is a drug used for the treatment of chronic bronchitis, bronchial asthma and chronic respiratory insufficiency, with precise efficacy and low toxic side effects. Currently, there are two main methods of synthesis of fenspiride: 1. 1-phenylethyl-4-piperidone is used as the starting material to produce 1-(2-phenylethyl)-4-hydroxy-piperidine by Reformatsky reaction. -4-Hydroxy-4-acetic acid ethyl piperidine, then Curtius rearrangement to obtain the product fenspiride, this method because of the Reformatsky reaction, so need anhydrous conditions, the reaction conditions require harsh, and at the same time to activate the zinc powd er This method requires anhydrous conditions and demanding reaction conditions, as well as activation of zinc powd er, which is cumbersome and dangerous to operate as the Curtius reaction is carried out to generate azide; 2. 4-piperidone is used as the starting material to produce 4-hydroxy-4-(1-nitro)methylpiperidine. Then hydrogen reduction of nitro to amino, and then react with phosgene to produce fenspiride, this method of reduction to pass hydrogen pressure reaction, hydrogen environment is prone to cause explosions, the production process is highly dangerous, and the subsequent use of phosgene is a highly toxic chemical gas, inconvenient to use in production, easy to cause environmental pollution.
